Did you read the section in the Gtk-OSX instructions about Lion, and how the old XCode3 SDKs don't work with XCode4?

Use the 10.6 SDK that came with XCode4.

You'll still encounter a problem at building Cairo; I'll have a patch for that up over the weekend.
