It doesn't really matter where you run it from as all of the paths are absolute, but you do want to have a shell in which you haven't sourced devrc.sh. 

Running install.sh to completion is a prerequisite for doing anything else, even compiling gnucash. It's the only way to make sure that all of the dependencies are correctly installed.
