If you want to completely separate your accounting from your wife's, the 
easiest would be to create to separate gnucash books (Via File->New) and 
follow the druid. In this way you can both track your expenses.

However, if you wish to keep track of some common assets (a common bank 
account for example), but you wish to keep track of both of your cash
expenses 
separately, you better create one gnucash book and under Assets define two 
cash assets: one for you and one for your wife.

No doubt there are still other possibilities, but your initial question is a

little vague.
