Yes, simply use "File->Open" to open (a copy of) your backup file.

It depends on the settings.

Some settings are global to your user account and hence will be seen by the 
new GnuCash version automatically and even carried over when saving your data 
to a database. This is generally the case for all settings under 
Edit->Preferences. 
Other settings are stored in your data file itself. These settings will be 
carried over when you save your data to a database.
Your custom reports will mostly be available as well. There were some changes 
from 2.2.x to 2.4.x in the report preferences though. As a result not all 
settings for all custom reports will be copied over. Unfortunately the budget 
report is one of them. See the related bugreport [1] for a workaround.
The same goes for reports that were left open the last time you closed 
GnuCash.
