No. Gnucash can work with both zipped or unzipped data files. 

A couple of gnucash versions had a bad version of the zip library and hence sometimes had issues reopening a badly saved data file. I don't remember whether manually unzipping and then reopening the unzipped file fixed this issue. It's certainly worth a try.
