GnuCash doesn't understand the signal from Finder about opening the file, so you have to use File>Open from the menu after starting GnuCash.

After selecting File>Open, you'll get a file system dialog box that works mostly like Finder. Pick the file-folder symbol with a / next to it in the sidebar, then scroll to the bottom of the list and double-click "Volumes". You should see your thumb drive listed, double click on it, then continue to navigate through its folders to where your .gnucash file is. 
After you've done that once GnuCash will open it every time on startup as long as you mount your thumb drive before you start GnuCash.
